Learning objective
Compare transmitter synthesis, clearance, receptor families and projection origins without treating one transmitter as one behavior.
Acetylcholine has its own recycling logic
Acetylcholine is synthesized from choline and acetyl-CoA. Acetylcholinesterase breaks it down in the cleft, and choline is taken up for reuse. This is not uptake of intact ACh. Nicotinic receptors are ionotropic; muscarinic receptors are GPCRs. Cholinergic projections participate in attention, arousal and memory, while peripheral cholinergic signaling has additional roles.
Catecholamines and serotonin
The catecholamine synthesis sequence is tyrosine → L-DOPA → dopamine → norepinephrine, with epinephrine a further product in appropriate cells. Serotonin is a monoamine derived from tryptophan, not a catecholamine. The course uses DA–DAT, NE–NET and 5-HT–SERT as canonical uptake associations; these are not universal claims of exclusive transporter substrates. MAO contributes to monoamine metabolism, and COMT contributes to catecholamine metabolism.
Origins, targets and receptor exceptions
Substantia nigra and VTA provide important dopamine origins. The locus coeruleus is a major norepinephrine origin with broad projections. Raphe nuclei are major serotonin origins. Dopamine and adrenergic receptor families are GPCRs. The serotonin exception to remember is 5-HT3, an ionotropic receptor; other discussed serotonin receptor families are GPCRs. Receptor subtype and circuit matter more than slogans such as “serotonin equals happiness.”
| System | Synthesis / recycling anchor | Receptor or pathway anchor |
|---|---|---|
| Acetylcholine | Choline + acetyl-CoA; AChE breakdown and choline reuse. | Nicotinic channel / muscarinic GPCR. |
| Dopamine | Tyrosine → L-DOPA → DA; DAT uptake. | Substantia nigra and VTA projections; D1/D2 families. |
| Norepinephrine | Formed from DA; NET uptake. | Locus coeruleus; alpha/beta adrenergic GPCRs. |
| Serotonin / 5-HT | Tryptophan precursor; SERT uptake. | Raphe nuclei; 5-HT3 ionotropic exception. |

Explain it without notes
Compare ACh clearance with serotonin clearance.
Compare your explanation 1
AChE breaks down ACh and choline is recycled. Serotonin can be taken up by SERT and later metabolized or packaged separately.
How do you distinguish NE and 5-HT on a pathway question?
Compare your explanation 2
Pair NE with locus coeruleus and NET; pair 5-HT with raphe nuclei and SERT. Then use the stated receptor and target rather than inferring an entire behavior.
